Apoptotic Death of Antigen-Carrying, Effete Promote Tolerance Induction by Eliciting Ligand-Expressing Invariant NK T Cells Oral Nickel Tolerance: Fas

چکیده
Whereas oral nickel administration to C57BL/6 mice (Ni high mice) renders the animals tolerant to immunization with NiCl 2 combined with H 2 O 2 as adjuvant, as determined by ear-swelling assay, it fails to tolerize J␣18 ؊/؊ mice, which lack invariant NKT (iNKT) cells. Our previous work also showed that Ni high splenic B cells can adoptively transfer the nickel tolerance to untreated (Ni low) recipients, but not to J␣18 ؊/؊ recipients. In this study, we report that oral nickel administration increased the nickel content of splenic Ni high B cells and up-regulated their Fas expression while down-regulating expression of bcl-2 and Bcl-x L , thus giving rise to an Ag-carrying, apoptosis-prone B cell phenotype. Although oral nickel up-regulated Fas expression on B cells of both wild-type Ni high and J␣18 ؊/؊ Ni high mice, only the former showed a reduced number of total B cells in spleen when compared with untreated, syngeneic mice, indicating that iNKT cells are involved in B cell homeostasis by eliciting apoptosis of effete B cells.

Induction of specific T cell tolerance by Fas ligand-expressing antigen-presenting cells.
Autocrine interaction of Fas and Fas ligand leads to apoptosis of activated T cells, a process that is critical for the maintenance of peripheral T cell tolerance. Paracrine interactions of Fas ligand with T cells also may play an important role in the maintenance of tolerance, as Fas ligand can create immune-privileged sites and prevent graft rejection by inducing apoptosis in T cells. We surm...
متن کاملExtinction of IL-12 signaling promotes Fas-mediated apoptosis of antigen-specific T cells.
In previous studies we have shown that peripheral tolerance achieved by high dose feeding of OVA to intact OVA-TCR transgenic mice was enhanced when endogenous IL-12 was neutralized simultaneously. To generalize this phenomenon, in the present study we investigated the tolerogenic mechanisms underlying the blockade of IL-12 signaling following oral and systemic Ag delivery. We found that the nu...
متن کاملAntiinflammatory Effects of CD95 Ligand (FasL)-induced Apoptosis
Apoptosis is critical to homeostasis of multicellular organisms. In immune privileged sites such as the eye, CD95 ligand (FasL)-induced apoptosis controls dangerous inflammatory reactions that can cause blindness. Recently, we demonstrated that apoptotic cell death of inflammatory cells was a prerequisite for the induction of immune deviation after antigen presentation in the eye. In this repor...
متن کاملUltraviolet light-induced immune tolerance is mediated via the Fas/Fas-ligand system.
Hapten sensitization through UV-exposed skin induces tolerance that is mediated via the induction of hapten-specific T suppressor cells. However, the detailed mechanisms underlying tolerance induction remain unclear to date. We show here that the apoptosis-related surface Ag Fas (APO-1, CD95) and its ligand, Fas ligand (FasL) are critically involved, since Fas-deficient lpr mice and FasL-defici...
متن کاملFas-mediated cytotoxicity by freshly isolated natural killer cells
The expression of Fas ligand on natural killer (NK) cells and Fas-mediated cytotoxicity by NK cells was investigated. Fas ligand mRNA was expressed in freshly isolated NK cells but not in T cells. Furthermore, the Fas ligand was detected on the cell surface of NK cells by staining with soluble Fas molecule. We analyzed the cytolytic activity of NK cells against thymocyte targets from normal and...
